Do you think humanity could spread into space if needed?

Let's say that an asteroid was going to hit the Earth and for some reason scientists are 100% sure that it will happen. If humanity had 1 year to prepare, do you think that we would be able to send enough people out to places like the moon or mars, to keep the species going. You would need roughly 250 breeding pairs (500 people in total). It would be the top priority of governments around the world. I feel that under these extreme conditions, humanity would be able to utilize already known technology like submarines to be able to make a self-sustaining colony, that would be able to grow and later colonize other places. What do you think?
